I don't consider the first two sentences natural, unless you have some specific context. I would say something like "How long are the working hours?"

The first sentence is asking how long work is. The second sentence is asking of all the hours, how many work are.
The first sentence is somewhat unnatural though I think, we would probably just ask "How long is work?" or "How many hours is work?"
The second sentence is natural and what "hours" is depends on the context.
